Angiopoietin-2, or ANGPT2, is known to be a ligand of the TIE2 receptor tyrosine kinase (TEK). Signaling of ANGPT2-TIE2 triggers vascular sprouting by working together with vascular endothelial growth factor, or VEGF.
ANGPT2 suppression exhibits prospective results in inhibiting tumor growth, tumor angiogenesis and metastasis in preclinical models. This makes it a promising candidate to target for cancer treatment.
Angiopoietins are a class of secreted factors that include ANGPT1, ANGPT2, ANGPT3 (ANGPTL1), ANGPT4, ANGPT5 (ANGPTL3), ANGPTL2, ANGPTL4, ANGPTL5, ANGPTL6 and ANGPTL7. While ANGPT1 is a potent activator of TIE2, ANGPT2 acts as a context-dependent agonist/antagonist.
During the early stages of tumor progression, ANGPT2 is highly upregulated to induce neovessel formation. Traditional anti-VEGF therapies mainly target immature blood vessels in tumors.
Currently, it is predicted that the combinational targeting of both VEGF and ANGPT2 pathways enhances clinical efficacy by widening the target vessel population. Sino Biological has come up with reagents of high quality to promote the study of ANGPT2/TIE2 signaling.
