In humans, there are two types of FcγR: the activating receptor, which contains the CD64 (FcγRI), CD32a (FcγRIIA), CD32c (FcγRIIC), CD16a (FcγRIIIA), and CD16b (FcγRIIIB); and the inhibiting receptor, CD32b (FcγRIIB). Table 1 lists the types and characteristics of FcγRs.

To mediate antibody-dependent cellular cytotoxicity (ADCC), NK cells recognize cell-bound antibodies via CD16A (FcγRIIIA). A new area of cancer immunotherapy is enhancing therapeutic mAb-mediated CD16 activation of NK cells.
CAR-NK cells containing multi-specific molecules made up of relatively stable CD16 and other activating receptors (e.g., NKG2D, DNAM-1/CD226, NKp46/NCR1, NKp44/NCR2, and NKp30/NCR3), scFvs for tumor antigens (e.g., CD19, CD22, ROBO1, and PSMA), and cytokine moieties (e.g. CD16 genetic modifications to eliminate sensitivity to ADAM-17-mediated proteolytic cleavage or increase affinity for IgG Fc region have shown promising results.
CAR-NK cells expressing CD16A in recombination with the high-affinity Fc receptor, CD64 (FcγRI), demonstrate a more stable and enhanced ADCC response, indicating a promising advancement in CAR-NK cell immunotherapy.

Figure 1 depicts the concept of using CAR-NK cells in combination with antibody immunotherapy to counteract NK cell impairment, in which CAR-NK cells can kill target cancer cells independently via CAR and also serve as CD16-positive effector cells for mitigating ADCC against cancer cells.
