Cellenion’s proteoCHIP product line offers scientists unique techniques for miniaturizing sample preparation of individual cells for single-cell proteomics workflows.
Recent advances in sample processing and mass spectrometry instruments have enabled protein characterization at the single-cell level. Individual cell proteome analysis enables the dissection of pathways that would otherwise go undetected. However, sample preparation methods still have the potential to unleash the full power of single-cell proteomics.
Cellenion has created customized methods to address difficulties in preparing single-cell proteomics samples.
For multiplex and label-free workflows, proteoCHIP solutions' ideal design and accessories enable automated and miniature sample preparation of individual cells before mass spectrometry analysis, further expanding the potential of cellenONE® single-cell dispensing technology.
All front-end mass spectrometry features, including incubation procedures, are made possible by these cutting-edge chips on the cellenONE®.

proteoCHIP 12*16 (multiplex workflow)

The proteoCHIP 12*16 is a cutting-edge multiplexed single-cell mass spectrometry-based proteomics analysis tool. It streamlines the entire front-end process on cellenONE, including critical incubation steps, ensuring seamless sample preparation for mass spectrometry.
The proteoCHIP 12*16 has a microscope slide footprint, enabling novel tiny single-cell proteomics applications in nanoliter volumes.
Each proteoCHIP has 12 arrays (pools) of 16 wells each, allowing for peptide labeling with TMTpro (11-plex or 16-plex). The cellenONE can process up to three chips per run.
After labeling, the cells can be conveniently pooled using the proteoCHIP Funnel. A specialized Accessory Kit includes all the adapters needed for proteoCHIP and Funnels with cellenONE, centrifuge, and LC-MS/MS autosamplers.
Benefits of proteoCHIP 12*16

- Standardization: Automated dispensing instead of manual pipetting improves repeatability
- Miniaturization: Nanoliter reaction volumes minimize reagent costs while increasing sensitivity
- Throughput: Prepare up to 576 single cells each run using cellenONE
- Process safety: Prefilling with oil avoids evaporation, while a special design and material avoid cross-contamination
- Recovery: Direct injection with an autosampler results in little peptide loss and maximum liquid recovery
- Data quality: With an exceptional signal-to-noise ratio, it can identify and measure up to 2000 proteins per single cell (even with low or no carrier)

The proteoCHIP LF 48 enables revolutionary miniaturized single-cell proteomics applications on a microscope slide footprint in nanoliter volumes. Each proteoCHIP can process up to 48 single cells, and up to three chips can be run simultaneously on cellenONE®, yielding up to 144 single cells each run.
The chips can then be directly interfaced with the LC-MS/MS system or manually placed into appropriate containers (e.g., Evotips).
Benefits of proteoCHIP LF 48
- Standardization: More reproducibility is made possible by automated dispensing rather than humans pipetting.
- Miniaturization: Sensitivity is increased by nanoliter reaction volumes
- Throughput: For each run on cellenON, prepare up to 144 single cells
- Zero vial exchanges: Direct connection to the autosampler
- Data quality: Identify and quantify over 1000 proteins per single cell
- Clustering and differentiation of cell types

proteoCHIP EVO 96 (label-free workflow with direct transfer to evotips)

Designed for robust and high-throughput liquid chromatography, single-cell proteomics has never been so easy.
proteoCHIP EVO 96 is a novel solution for label-free single-cell proteomics sample preparation. Its excellent design and accessories enhance the possibilities of cellenONE® single-cell isolation technology. The combination of these technologies allows for the complete creation of miniaturized single-cell proteomics samples for MS-based analysis.
Using the proteoCHIP EVO 96, single-cell samples can be prepared directly on the cellenONE deck, including incubation. After sample preparation, centrifugation can transfer samples straight into Evotips, eliminating the need for a manual step.
Testimonials
I am using the proteoCHIP EVO 96 for very diverse applications, including single-cell proteomics and spatial tissue profiling. The proteoCHIP EVO 96 streamlines our label-free single-cell protocol to only a couple of hours with even less sample handling compared to the previous multiplexed proteoCHIP. One of the biggest advantages to perform sample preparation on the EVO 96 is the confident cell deposition in the sample droplet and the minimal evaporation during processing. The direct connection of the chip to the Evosep additionally allows us to clean up the samples with minimal losses, protect the column, and reproducibly acquire 80SPD at biologically relevant proteome depth."
