The TREM (triggering receptor expressed on myeloid cells) family is a collection of cell surface receptors. This family has garnered much interest because of its ability to fine-tune the inflammatory response. TREM proteins, particularly TREM-1 and TREM-2, are presently being investigated as potential therapeutic targets for a variety of disorders, including neurological diseases, inflammatory diseases, and malignancies.

TREM-1, TREM-2, TREM-3, TREM-4/CLM-9/CD300LG, TREM-5/CLM-7/CD300LB, plasmacytoid dendritic cell-TREM (PDC-TREM), and TREM-like receptors TREML1/TLT-1 and TREML2/TLT-2 are all members of the TREM family.

TREM protein ligands have been elusive so far. TREM1 has only been linked to the high mobility group box 1 (HMGB1), extracellular actin, peptidoglycan recognition protein 1 (PGLYRP1), and cold-inducible RNA-binding protein (CIRP).

TREM2 ligand Amyloid, which is well-studied, is a crucial pathogenic protein in Alzheimer’s disease. B7-H3, a well-studied TLT-2 ligand, is essential for adaptive immune responses. The functional role of TREM ligands in diseases strengthens the case for TREM proteins as promising medicinal targets.
