How to optimize nuclei preparation for better genomic results

Precisely counting nuclei and intact cells is crucial in preparing for future applications. Miltenyi Biotec researchers used a flow cytometer and a DeNovix CellDrop Automated Cell Counter to count nuclei.

Nuclei samples counted on the CellDrop (FL or FLi) were treated with Acridine Orange and Propidium Iodide fluorescent dyes to distinguish between isolated nuclei, entire cells, and debris.

The variable protocol choices and robust analysis method in CellDrop improved the accuracy of the nuclei count results. The study found that CellDrop's count results were close to those of the Miltenyi Biotec MACSQuant Analyzer 10 Flow Cytometer, indicating great accuracy in both approaches.
