Simoa® Bead Homebrew Assay
The Simoa® Bead Homebrew Assay can be used with HD-1/HD-X and SR-X Systems. This innovative solution presents unparalleled surface chemistry. Bead-based immunoassays can also advance your research capabilities, starting with capturing and labeling analyte using the enzyme conjugate streptavidin ß‐galactosidase (SBG).
The analyte is captured with beads, and a biotinylated detection antibody is then added to create a captured analyte immunosandwich. The immunosandwich is labeled with SBG, with washing and incubation steps tailored to your personalized assay protocol.
A magnet pelletizes beads after every incubation. After a final wash cycle, the capture beads are resuspended in resorufin ß‐D‐galactopyranoside (RGP) substrate, ready for imaging and analyte quantification on a Simoa® disc.
Simoa® Planar Homebrew Assay
The Simoa® Planar Array Homebrew Assay used with the SP-X Imaging and Analysis System™ provides universal assay plates with immobilized anchor antibodies in 96 microwells. These are very specific for use with the Simoa® planar arrays.
Everything required to perform peptide tag labeling for up to 6 capture antibodies and biotinylation for up to 6 detector antibodies is provided in the kit.
In the Simoa® planar assay format, the peptide-tagged capture antibody initially binds the plate-bound anchor. The assay then follows the familiar sandwich immunoassay format, with the antigen sandwiched between the peptide-tagged capture.
Biotinylated detector antibodies, streptavidin-HRP, and a chemiluminescent HRP substrate work together to signal the formation of immunocomplexes. The intensity of this signal directly correlates with the quantity of the analyte present in the standard or sample of interest.
