Tight junctions are a type of cell-to-cell adhesion found in epithelial and endothelial cell layers. They form continuous seals around cells, acting as a barrier to prevent the free passage of solutes and water through the spaces between cells.
These junctions consist of sets of continuous networking strands on the outer cytoplasmic leaflet, with matching grooves on the inner extracytoplasmic leaflet. Claudin-6 (CLDN6) is a multipass transmembrane protein belonging to the Claudin family. It is expressed by epithelial cells and plays a role in tissue development and maintaining the integrity of tight junctions.
CLDN6 is also one of the entry cofactors for the hepatitis C virus. Additionally, the methylation of CLDN6 may contribute to the development of esophageal tumors. The CLDN6 gene is located next to another Claudin family member, CLDN9, on chromosome 16.

CL6-HF2M9-Cell-based assay. Image Credit: ACROBiosystems Inc.
200,000 anti-Claudin-6 CAR-293 cells were stained with 100 μL of 1 μg/mL Fluorescent Human Claudin-6 Protein, Flag, His, GFP Tag (Cat. No. CL6-HF2M9) and a negative control protein, respectively (Fig. A and B). The FITC signal was used to assess the binding activity, and this was quality control tested.

The Human Claudin-6 Full Length Protein (VLP) (Cat. No. CL6-H52P7) captured on an L1 Chip can bind to the Anti-Claudin-6 Antibody, Human IgG1, with an affinity constant of 57.8 pM, as determined by an SPR assay (Biacore 8K). This is routinely tested.
CL6-HF2G8-SPR. Image Credit: ACROBiosystems Inc.
The Fluorescent Human Claudin-6 Full Length Protein (VLP) (Cat. No. CL6-HF2G8) captured on an L1 Chip can bind to the Anti-Claudin-6 Antibody, Human IgG1, with an affinity constant of 68.7 pM, as determined by an SPR assay (Biacore 8K). This binding interaction is routinely tested.
