Synuclein alpha increases local Ca2+ release from microdomains, which is crucial for the enhancement of ATP-induced exocytosis.

In its multimeric membrane-bound state, the protein acts as a molecular chaperone, assisting in the folding of synaptic fusion components known as SNAREs (Soluble NSF Attachment Protein REceptors) at the presynaptic plasma membrane, in conjunction with cysteine string protein-alpha (DNAJC5). This chaperone activity is vital for maintaining normal SNARE-complex assembly during aging. Additionally, the protein regulates dopamine neurotransmission by associating with the dopamine transporter (DAT1) and modulating its activity.
