Interleukin-17 (IL17): A pro-inflammatory cytokine that drives autoimmune diseases

T helper 17 (Th17) cells generate the pro-inflammatory cytokine interleukin-17 (IL17/IL17A). IL17 has been a target for a variety of therapeutic monoclonal antibodies and inhibitors to treat different inflammatory diseases and cancers due to its vital functions in immune control.

IL17 is part of a cytokine family that includes five other members: IL17B, IL17C, IL17D, IL17E/IL25 and IL17F. The sequences of IL17 and IL17F are the most similar. To signal through the receptor complex constituted of the subunits of IL17RA and IL17RC, IL17 and IL17F form homodimers and heterodimers (IL17A & IL17F). Both IL17 and IL17F have a role in attracting neutrophils to inflammatory areas.

The production of cytokines (IL-1β, TNFα, G-CSF, IL-6, etc.), chemokines (MCP-1/CCL2, CXCL1, CXCL2, CCL20, etc.), inflammatory effectors and antimicrobial proteins can be induced by IL17 and IL17F signaling.

At least two more ligands, IL17C and IL17E, utilize the IL17RA subunit. IL17RB, IL17RC, IL17RD and IL17RE are all members of the IL17 Receptor family.
