Stem Cell Factor (SCF), also known as kit-ligand (KL), steel factor, KITLG, FPH2, KL-1, Kitl, MGF, SF, or SHEP7, is a cytokine that binds to the c-Kit receptor (CD117). SCF exists in both transmembrane and soluble forms, generated through alternative splicing of the same RNA transcript. It is crucial in hematopoiesis (blood cell formation), spermatogenesis, and melanogenesis.
Fibroblasts and endothelial cells produce forms of SCF, with the soluble version having a molecular weight of 18.5 kDa and forming a dimer. SCF is essential for hematopoiesis during embryonic development, with key hematopoietic sites such as the fetal liver and bone marrow expressing this cytokine. SCF also influences the localization of melanocytes, the pigment-producing cells responsible for skin and hair coloration.
In the adult bone marrow, SCF plays a regulatory role in the hematopoietic stem cell (HSC) niche. It is often combined with other cytokines to culture HSCs and hematopoietic progenitors. The ability to expand these cells ex vivo presents significant potential for advancements in bone marrow transplantation, enabling the re-establishment of blood cell formation in patients.
