FGF basic is a member of the FGF family, which includes at least 23 related mitogenic proteins sharing 35–60% amino acid conservation. Unlike most other FGFs, FGF acidic and FGF basic lack signal peptides and are secreted through non-classical pathways.
FGF basic has been isolated from various sources, including neural tissue, pituitary, adrenal cortex, corpus luteum, and placenta. This factor contains four cysteine residues; however, its biological activity remains intact even in its reduced form, indicating that disulfide bonds are not essential for function.
bFGF is a key component of human embryonic stem cell culture media, required to maintain cells in an undifferentiated state, though the exact mechanisms remain unclear. It has been shown to induce gremlin expression, which inhibits differentiation triggered by bone morphogenetic proteins. bFGF is necessary in both mouse feeder cell-dependent culture systems and feeder- and serum-free culture conditions.
