FCγ receptors (FCγRII/CD32, FCγRIII/CD16, and FCγRI/CD64) are known as cellular receptors for IgG that add up to immune complex clearance, antibody-dependent cell-mediated cytotoxicity (ADCC), phagocytosis, and comprise several subtypes.

FCγR polymorphisms can potentially forecast responses to immunotherapies, thereby directing personalized treatment methods. As far as therapeutic antibody development is concerned, the efficacy of mAbs is identified by their affinity to FCγRs, which impacts their engagement with immune effector cells.

FcγR-binding is vital for developing immune checkpoint antibodies against CTLA-4, PD-1, TIM-3, PD-L1, LAG3, CD73, TIGIT, VISTA, and B7-H3. Interactions of mAb with FcγRs also affect cytokine release, which is vital in the immune response. Moreover, FCγRs can serve as biomarkers to monitor treatment responses and are studied in combination therapies such as HER2-targeted mAb therapy.
