Interleukin-18 (IL-18) is a pro-inflammatory cytokine with several functions. It is a member of the IL-1 superfamily and is involved in both innate and acquired immunity. The IL-18 pathway has been linked to autoimmune illnesses, metabolic problems, and malignancies in recent investigations.
In collaboration with IL-12, IL-18 regulates host immunity by modulating IFN-production. IL-18 stimulates natural killer (NK) cells, natural killer T (NKT) cells, and established Th1 cells to generate IL-3, IL-9, and IL-13 in the presence of IL-2.
IL-18 promotes downstream signal transduction in the same way as IL-1β does. IL18 interacts with the IL-18 receptor α. The downstream cellular signal is initiated once IL-18Rβ/ IL-18RAcP is recruited to the complex. IL-18 signaling is inhibited by IL-18BP, a secreted antagonist receptor. In conjunction with SIGIRR, IL-18Rβ may bind IL-37 and transmit an anti-inflammatory signal.

Recombinant IL-18 receptors
The IL-18 receptor complex is a heterodimer composed of a (IL-18Rα) chain that is responsible for IL-18 extracellular binding and a nonbinding, signal-transducing β (IL-18R β/IL-18RAcP) chain.
