Assessing peptide stability under thermal stress conditions

Study: Comparing peptide stability under similar stressors

Two peptide samples from distinct production lots were exposed to comparable stressors to identify which had a higher propensity for aggregation and inherent instability.

Method and stressors:

The researchers prepared six samples of each peptide lot at 1mg/ml in two pH solutions, 2.5 and 3.0. Three of the six solutions were next exposed to thermal stress of 18C, 25C and 35C alone while the remaining samples were exposed to thermal stress and stirring stress at 300RPM.

Analysis:

The following graph demonstrates the quantified Aggregation Rates for all peptides with and without stirring and at various temperatures and pH levels. The data suggests that peptide Lot 1 is more stable than Lot 2 when exposed to conditions of thermal and stirring stress. Changing the solution pH from 3.0 to 2.5 induced an extra stability gain of around 10x reduction in the AR value.
