The Saliva DNA Collection and Preservation Devices from Norgen are made to 1) collect saliva in a straightforward and non-invasive manner and 2) preserve DNA in saliva samples at room temperature. There are three parts to each of the 50 saliva DNA collection and preservation devices:

• Saliva Collection Funnel and Collection Tube
• Collection Tube Cap
• A sealed, squeezable ampoule containing Norgen's Saliva DNA Preservative

Saliva samples are collected by spitting into the Collection Funnel, which is pre-assembled with the Collection Tube. Once the required volume of saliva is collected, the funnel is removed and the contents of the Preservative Ampoule are added to the tube and mixed thoroughly with the saliva. The sealed Saliva Collection Tube is then ready to be sent to the lab for DNA isolation and analysis.

DNA can be extracted from the preserved samples using Norgen’s Saliva DNA Isolation Kit (Cat. RU45400). Each Collection Tube is marked with a unique serial number, enabling secure and anonymous tracking of individual samples.

The preserved saliva DNA remains stable at room temperature for over two years, making this kit a reliable option for long-term storage. It’s especially well-suited for large-scale DNA collection in epidemiological and population studies.

Saliva DNA preservative

Norgen's Saliva DNA Preservative is an aqueous storage buffer formulated to enable rapid cellular lysis and long-term preservation of DNA from fresh saliva specimens. It’s designed to prevent the growth of Gram-negative and Gram-positive bacteria, as well as fungi, and also inactivates viruses—rendering samples non-infectious and safe for handling and transport.

One of the key benefits of this buffer is that it removes the need for immediate processing or freezing. Samples can be safely shipped to centralized testing facilities at room temperature, simplifying logistics and reducing costs. The buffer’s formulation ensures that DNA remains stable, with no detectable degradation, for more than two years.

DNA isolation from preservative

Before beginning saliva DNA isolation, vortex the Collection Tube containing the preserved saliva for 10 seconds, then incubate it at 55 °C for one hour. Saliva DNA can now be isolated from the preserved saliva samples using any commercially available method, including Norgen’s Saliva DNA Isolation Kit (Cat. RU45400), Norgen’s Saliva DNA Isolation 96-Well Kit (Cat. RU35200) and Norgen’s Saliva DNA Isolation Kit (Magnetic Bead System) (Cat. RU55400, RU62900). 

Norgen’s Saliva DNA Isolation Reagent Kit (up to 4 mL) (Cat. RU35720) also allows for DNA isolation from the preserved saliva samples using an alcohol precipitation method. The purified DNA is of the highest quality and can be used in a number of downstream applications, including PCR, NGS, and microarray analysis.

Figure 1. Stability of DNA Preserved in Norgen's Saliva DNA Preservative at Room Temperature for over 2 Years. Saliva samples were collected from numerous donors and mixed, and then an equal volume of Norgen's Saliva DNA Preservative was added to the saliva. The preserved saliva was then stored at room temperature for up to 24 months. Saliva DNA was subsequently isolated at 4 months, 8 months, 16 months, and 24 months from 0.5 mL of the saliva/preservative sample using Norgen's Saliva DNA Isolation Kit (Cat. RU45400). For visual analysis, 100 µL of the purified DNA was run on an agarose TAE gel. As it can be seen, there is no evidence of DNA degradation after the saliva samples are stored for 24 months at ambient temperatures in Norgen's Saliva DNA Preservative. Shelf life and handling

• Unused Norgen Saliva Preservative Tubes are stable through to the collect-before date without any reduction in performance when stored at room temperature. See the device insert or tube label for the collect-before date.
• After its collection, saliva is then stable for over two years when kept tightly sealed and stored at room temperature.
• The Collection Tube, Funnel, and Container of each Individual Saliva DNA Collection and Preservation Device are recyclable.
