Overview
Features and benefits
- In less than 30 minutes, intact exosomes were purified and enriched from plasma, serum, urine, cell culture media, and saliva
- Sample input ranges from 5 to 35 mL
- Cell Culture Media Exosome Purification Mini Kit (5 mL - 10 mL)
- Cell Culture Media Exosome Purification Maxi Kit (20 mL - 35 mL)
- Exosome purification relies on Norgen’s proprietary resin
- Protease or coagulant treatments, overnight incubation, or precipitation reagents are not necessary
- No need for time-consuming filtration, ultracentrifugation, or specialized syringes
- Depending on the type of sample input, purify intact exosomes that range in size from 40 to 200 nm
- Functional studies can be conducted with purified exosomes
- Purified exosomes are free from any other RNA-binding proteins
- Purified exosomes can be assessed using NanoSight® or Electron Microscopy to determine their approximate size range and concentration
- Exosomal RNA can be recovered from purified exosomes utilizing Norgen’s Exosomal RNA Purification technology or any other RNA extraction method
The Cell Culture Media Exosome Purification Kits offer a quick, dependable, and convenient way to purify and enrich intact exosomes from various cell culture media sample sizes ranging from 5 mL to 35 mL. These kits also enable the purification of intact extracellular vesicles (EVs) from various cell culture media sample quantities, resulting in EVs suitable for the majority of downstream applications. The purification is based on Norgen's proprietary resin.
These kits have a distinct benefit over other approaches because they do not require any specialized equipment, ultracentrifugation, precipitation reagents, or protease treatments. More significantly, the purified exosomes will not contain any other RNA-binding proteins that could contaminate the exosomal RNA, which is critical when investigating exosomal transcripts.
NanoSight® analysis
NanoSight® can be used to evaluate exosomes enhanced with Norgen's Cell Culture Media Exosome Purification Kits to determine their approximate size range and concentration.
Exosomal RNA analysis
Use the Cell culture media Exosome Purification and RNA Isolation kits to purify exosomes and isolate exosomal RNA. The methodology is separated into two phases, with an aliquot of pure exosomes available for applications such as NTA/TEM before being processed for RNA isolation.
Users can also use the Exosomal RNA separation kit if exosomes are previously purified with a Norgen kit or another approach. Exosomal RNA isolation is based on Norgen’s unique resin, eliminating the requirement for phenol extractions or carrier RNA. This RNA is appropriate for gene expression analysis (RT-qPCR, microarray, or NGS) and biomarker development.

Figure 1. Isolation of RNA from exosomes purified from different cell culture media volumes. Norgen's Cell Culture Media Exosome Purification Mini Kit (Cat. 60400) was used to isolate exosomal RNA from exosomes purified from different cell culture media volumes using the same kit. 2 µL of the isolated RNA was then used as the template in RT-qPCR reactions to assess the amplification of the isolated exosomal RNA (A) The exosomal avg. Ct value for miR-26a is linearly decreasing with increasing the sample input volume. (B) The relative amount of the exosomal miR-26a shows excellent linearity with a percentage of recovery of more than 90%. Image Credit: Norgen Biotek Corp.

Norgen's Cell Culture Media Exosome Purification Maxi Kit (Cat. 60600) was used to isolate exosomal RNA from different cell culture media volumes from exosomes purified using the same kit. 2 µL of the isolated RNA was then used as the template in RT-qPCR reactions to assess the amplification of the isolated exosomal RNA.
