Fibronectin (Fn) is a glycoprotein ranging from 230 to 270 kDa, typically existing as a dimer linked by disulfide bonds at the C-termini. Each monomer consists of three repeating units: 12 Type I, 2 Type II, and 15–17 Type III domains, collectively making up about 90 % of the Fn sequence.
The extracellular matrix (ECM) functions as both a structural scaffold and a regulator of cell signal transduction within tissues. Fibronectin plays a crucial role in the trabecular meshwork (TM), where it is found in the sheath material surrounding elastin tendons that extend from the ciliary muscle within the ciliary body.
During ECM assembly and turnover, cells increase Fn production, integrating it into viscoelastic fibrils capable of binding more than 40 distinct growth factors and cytokines. These fibrils are essential for provisional ECM formation in embryonic development and wound healing. Additionally, fibril assembly is upregulated in pathological conditions such as cancer and fibrotic diseases.
