Interleukin-6 (IL-6), also known as HGF, BSF-2, HSF, IFN-β2, and IL-6, was initially identified as a B-cell differentiation factor. It is a multifunctional cytokine that regulates immune responses, hematopoiesis, acute phase reactions, and inflammation. IL-6 is secreted by various cell types, including T cells, macrophages, monocytes, fibroblasts, and endothelial cells, primarily in response to trauma—such as burns or tissue damage—triggering inflammation.

IL-6 interacts with the interleukin-6 receptor and glycoproteins, playing a key role in several disease processes, including diabetes, atherosclerosis, depression, Alzheimer's disease, systemic lupus erythematosus, prostate cancer, and rheumatoid arthritis. Notably, patients with advanced or metastatic cancer often have elevated IL-6 levels in their blood. This has led to significant interest in developing anti-IL-6 therapies to target these conditions.
