Laminin 511 E8 (LN511-E8) is a recombinant human protein that offers a defined surface for feeder-free culture of various human pluripotent stem cells (PSCs) in vitro. As a truncated version of laminin 511, LN511-E8 is a functionally minimal form that retains the full ability to bind to integrins.
Without inducing differentiation, LN511-E8 has been shown to sustain normal growth characteristics and preserve stemness in multiple PSC lines, including ESC, iPSC, and MSC. Furthermore, LN511-E8 has demonstrated the ability to support PSC growth for over 10 passages without any signs of karyotypic abnormalities while maintaining the PSCs' capacity to differentiate into all three germ layers.
As Takamichi Miyazaki et al. reported, the LN511-E8 variant of laminin 511 demonstrates higher efficiency in supporting the adhesion of dissociated cells compared to wild-type laminin 511, making it a more cost-effective choice.

Laminin 511 E8 (LN511-E8) has been shown to support single-cell seeding at low density. The seeded cells exhibit high motility and improved clonal survival in a feeder-free culture system.

The laminin 511 isoform plays a crucial role in the growth and maintenance of human pluripotent stem cells (hPSCs) through its interaction with the a6B1 integrin receptors. LN511-E8, a truncated isoform of laminin 511, is effective across various stem cell lines, including iPSC, hESC, and MSC.
Better adhesion
The LN511-E8 variant of laminin 511 is more efficient than wild-type laminin 511 in supporting the adhesion of dissociated cells.
