Interleukin-3 (IL-3) is a human protein encoded by the IL3 gene. IL-3 plays a key role in the immune system as a cytokine, enhancing the body's natural response to disease. It exerts its effects by binding to the interleukin-3 receptor, which consists of a specific α-chain and a shared β-chain (βc). GM-CSF and IL-5 also utilize the latter. In some cases, IL-3 can bind to an alternative βIL-3 chain, which appears unique to IL-3.

Upon receptor binding, IL-3 activates Jak2, phosphorylating multiple Stat proteins and activating the PI3K/Akt signaling pathway. This signaling cascade contributes to airway inflammation, particularly in asthma.

The human IL3 gene is located on chromosome 5, just 9 kilobases from the GM-CSF gene, reflecting its functional similarity to GM-CSF. The IL-3 protein is 152 amino acids long and naturally glycosylated.
