Interleukin-4 (IL-4) is a cytokine differentiating naive helper T cells (Th0) into Th2 cells. Along with IL-13, IL-4 plays a key role in Th2-type immune responses, particularly in allergic reactions and parasitic infections. In the presence of these cytokines, macrophages undergo differential activation.
IL-4 is a ligand for the interleukin-4 receptor (IL-4R), which also binds IL-13, leading to overlapping functions between these two cytokines. The signal transducer and activator of transcription 6 (STAT6) has been identified as a crucial mediator of IL-4’s immune regulatory effects.
Recent research has highlighted IL-4’s role in developing innate CD8+ T cells in the thymus, particularly in gene-deficient mouse models lacking Itk, KLF2, CBP, and Id3. This process occurs without prior antigen exposure, suggesting IL-4’s involvement in shaping innate immune responses.
