VEGF165 is the most abundant splice variant of VEGF-A and is produced by various cell types, including endothelial cells, macrophages, and T cells. It plays a critical role in angiogenesis, promoting vascular endothelial cell survival, growth, migration, and permeability.

Hypoxia, inflammatory cytokines, and oncogenes regulate VEGF expression. VEGF165 specifically binds to heparan sulfate, allowing it to be retained on the cell surface and within the extracellular matrix. It interacts with the receptor tyrosine kinases VEGFR1 and VEGFR2, as well as co-receptors NRP-1 and NRP-2, which enhance VEGFR2 signaling.

VEGF165 activates VEGFR1 and VEGFR2 and triggers key signaling pathways, including PI3K/AKT, p38 MAPK, FAK, and paxillin. These pathways contribute to VEGF’s crucial role in tumor angiogenesis across various cancers.

There is no “tag” on this protein.

The protein’s estimated MW is 19.2 kDa. When measured against the Star Ribbon Pre-stained Protein Marker under reducing (R) conditions (SDS-PAGE), the protein migrates as 24 kDa ± 3 kDa as a result of glycosylation.

Endotoxin

The LAL technique yields less than 10 EU/mg.

Host cell protein

<0.5 ng/µg of protein tested by ELISA.

Host cell DNA

<0.02 ng/μg of protein tested by qPCR.

Sterility

Membrane filtration was used for sterility testing, as per CP<1101>, USP<71>, and Eur. Ph. 2.6.1 protocols.
